The development of a nanocrystalline apatite reinforced crosslinked hyaluronic acid-N,O-carboxymethylchitosan (NOCC) composite as an injectable bone cement

摘要

In this study, a water-soluble chitosan derivative, N,O- carboxymethylchitosan (NOCC) has been synthesized. Hyaluronic acid (HA) could evolve into a better cross-linked hydrogel by conjugating with NOCC, using carbodiimide as a cross-linker, therefore decreased the swelling capability and increased the resistance against hyaluronidase degradation of the HA-based hydrogels. The NOCC/HA mixed solution, after reacting with carbodiimide, increased its viscosity instantously and formed a hydrogel having a relatively low degree of swelling and high resistance against hyaluronidase. An injectable bone cement composed of nanocrystalline hydroxyapatite (HAp) and crosslinked NOCC/HA were developed by directly blending apatites with the NOCC/HA mixed solution, followed by crosslinked with carbodiimide. The mechanical strength of the NOCC/HA hydrogels were tuned by varying the apatite loading and NOCC/HA mixed ratio. These results indicated that the nanocrystalline HAp reinforced NOCC/HA composites make them attractive for as an injectable bone cement.
