Alzheimer’s disease is the most common cause of dementia characterized by progressive decline in memory and other aspect of cognition in the elderly. Although there is no treatment at present, early diagnosis of Alzheimer’s disease is important for the current drug therapies. Currently, Aricept, Exelon and Reminyl are only used to improve memory or delay memory loss in people with mild to moderate Alzheimer’s disease. However, accurate diagnosis of Alzheimer’s disease is sometimes difficult due to lack of reliable detection methods. Therefore, the development of the reliable and sensitive method for the early detection of Alzheimer’s disease is the most important issue for the improvement of the treatment efficiency. Recently, the biomarker can be used as an indicator for objective measurement and evaluation of normal biological processes, pathogenic processes, pharmacologic responses to therapeutic intervention and even to assess the risk or the presence of a disease. Here, we intend to design a rapid, reliable and sensitive platform to detect the biomarkers for the early diagnosis of Alzheimer’s disease. In this proposal, we plan to prepare substrates by silver nanocrystals for the detections of Alzheimer’s disease biomarkers in surface-enhanced Raman spectroscopy. To solve the issue of the low sensitivity of Raman, we try to design the surface-enhanced Raman scattering-based substrate to increase the signal intensity of Raman spectroscopy. Recently, we have synthesized monodisperse colloidal solutions of silver nanocrystals with regular polyhedral shapes such as cube, cuboctahedral and octahedral by simple polyol method. The as-prepared silver nanocrystals exhibited a broad surface plasmon resonance absorption band from the visible into the ultraviolet with a high absorption coefficient and can be used to prepare the substrate with the property of surface-enhanced Raman scattering. After successful preparation of silver nanocrystals, the homogenous, flat and clean two-dimensional silver nanocrystals Langmuir-Blodgett film will be prepared and served as the substrate in surface-enhanced Raman spectroscopy. Afterward, the surface of the substrate will conjugated with antibodies of Alzheimer’s disease biomarkers for the rapid screening test of Alzheimer’s disease in the early stage by surface-enhanced Raman spectroscopy. Eventually, we hope that we will be able to develop a rapid, sensitive and reliable detection platform using silver nanocrystal Langmuir-Blodgett films as the substrate for the detections of different disease biomarkers by surface-enhanced Raman spectroscopy.
