Design and Evaluation of a Novel Drug-Containing Ceramic Hydrogel for the Healing of Alveolar Bone Defect

Artificial bone scaffolds are widely used in current oral surgeries. Traditional bone scaffolds are designed mainly for the attachment and growth of osteoblasts, however, dental stem cells are also important and should not be ignored in the healing of mandibular bone defect. This study is devoted to developing a novel bone graft that has two advanced properties: (1) chemoattractive to dental mesenchymal stem cells (DPSC); (2) suitable for the attachment and growth of DPSC and osteoblast; (3) osteogenic inducing for DPSC and osteoblast. For this purpose, several chemokines candidates will be screened for their ability to chemoattract DPSC. In addition, flavonoids that could promote osteogenesis of DPSC and human primary osteoblast are selected. The flexible ceramic hydrogel scaffold is constituted with hydrogel and nano-hydroxyapatite for the attachment and growth of cells. In in vitro study, the drug-containing ceramic hydrogel will be evaluated in DPSC cells and human primary osteoblasts. In addition, the swine animal model will be applied for the evaluation of this ceramic hydrogel in vivo in the healing of mandibular bone defect. This study is important for the development of an advanced artificial bone graft in the future.
