本研究擬探討高度糖化終產物及其前驅物甲基乙二醛誘發腎臟與內皮功能損傷之負 面效應,並釐清該損傷機轉是否與活化發炎體有關;此外,另探討白藜蘆醇之介入是否對 於上述損傷具有保護作用。第一部分實驗以高度糖化終產物接受器抑制劑釐清腎臟中高度 糖化終產物接受器的表現是否和發炎體路徑的活化有關,此外評估不同種高度糖化終產物 對於小鼠腎功能之影響。第二部分實驗則以高度糖化終產物接受器抑制劑證實抑制受器和 高度糖化中產物之交互作用是否可達到減少自由基產生進而抑制發炎體的活化以減緩發 炎反應所造成的腎損傷。第三部實驗分則觀察白藜蘆醇對於以高度糖化終產物前驅物MG 誘發腎損傷之影響並釐清白藜蘆醇是否可透過影響發炎體的活化達到延緩腎臟受損的作 用。研究預期長期高度糖化終產物和其前驅物‐甲基乙二醇的投與會使高度糖化終產堆積 在腎臟,也可能經由與其接受器結合進入細胞中活化發炎體路徑使腎臟發炎,使腎臟和內 皮功能損傷。因此減少暴露於高度糖化終產物的環境下或是抑制其接受器的作用皆可抑制 發炎體的活化達到延緩腎損傷的作用,且白藜蘆醇的介入對於上述損傷可能具有保護作用。

Description

The aim of the study is to investigate the effects of resveratrol on advanced glycation end product and methylglyoxal-induced renal and endothelial injury. In the first part of the study, we will investigate the influence of receptor for advanced glycation end product on inflammasome pathway and we will also evaluate the effects of different advanced glycation end products on renal function in mice. In the second part, we will clarify the inhibition of inflammasome activation via blocking advanced glycation end product-receptor interaction through decreasing oxidative stress. In the third part, we will further investigate the beneficial effects of dietary resveratrol supplementation on methylglyoxal-induced renal injury via inhibition of inflammasome activation. We expect resveratrol may show its protective effects on advanced glycation end product and methylglyoxal-induced renal injury by regulating the inflammasome related inflammatory responses.
